4. Complete and describe the scheme of culture and identification  of meningococcal infections

Culture: blood or CSF is cultured on chocolate and blood agar in an atmosphere containing 5% of CO2 at 370C for 48 hours. Oxidase test is performed on colonies on solid medium. Sugar utilization test or commercial kits are used to identify meningococci.

Specimen:    
1 stage     Staining by ________________  

Inoculation on enriched selective medium or serum agar

 

2 stage     Oxidase test    
3 stage   Subculture in differential media for biochemical properties   Culture identification based on sugar fermentation and serological identification of the meningococcal capsule antigen by IF, ELISA and serovars
  1. Non-culturable methods:

- Capsule polysaccharide demonstration in CSF;

- Latex-agglutination test;

- PCR-based techniques.

 

  1. List pathogenic factors of:

Meningococci                                                            Gonococci

7. Describe preparations that can be used for treatment, diagnosis and prophylaxis of gonococcal and meningococcal infections: (antibiotics, vaccines, diagnostic meningococcal sera, ELISA test-systems).
